Известия о храме Успения Пречистой Богородицы в селе Кузьминском, вотчине Высоцкого монастыря, имеются под 1552 г. Около 1627 г. в местный приход входили деревни Рыжиково, Петровское, Верхние и Нижние Велеми. К 1694 г. на месте прежнего деревянного был возведен каменный храм: «сентября в 17 день по благословенной грамоте, дан антиминс ко освящению церкви Успения Пресвятой Богородицы в Серпуховской уезд в село Кузминское, взял антиминс той ж церкви поп Василий».
В XVII-XVIII в. в Успенском храме служили представители священнической династии Никифоровых. В 1705 г. из села выделилась деревня Новые Кузьменки на большой Московской дороге.
В 1924 г. в село Кузьменки переехал преосвященный Арсений (Жадановский), возглавлявший в 1914-1923 гг. Серпуховскую епископскую кафедру и расстрелянный в Бутове 27 сентября 1937 г. В половине дома, предоставленной владыке настоятелем храма протоиереем Михаилом, была устроена домовая церковь, в которой служил епископ. В советское время храм с. Кузьменки был единственным действующим сельским храмом во всем Серпуховском районе.
