旅籠紀伊国屋は東海道31番目の宿場「新居宿」にある。二十数軒あった宿の中でも最大の規模だったらしく、また経営者が紀州出身であったことから紀州藩の御用宿でもあった。江戸時代の建物は平屋建てだったのだが火災にあい、明治初期に二階建ての建物として再建され、現在に至っている。明治期の建物だが随所に江戸期の建築様式を色濃く残している。
宿としては昭和三十年代に廃業したが、その後は資料館として整備、公開されている。
